743316 Integrative Freshwater Ecology
summer semester 2019 | Last update: 13.11.2023 | Place course on memo listStudents will have hands-on in field (Research Station Gossenköllesee) and laboratory work related to the lecture. The students will acquire detailed knowledge on different methods (HPLC, underwater radiometry, organic carbon analyzer, spectrophotometry) used to characterize the dissolved organic matter, underwater optical properties, effects of solar radiation and photoprotective compounds by doing measurements and experiments.
The tutorial includes field and laboratory measurements/experiments on:
1) Sources of artificial UV radiation and materials used in experiments to test the effects of UV radiation
2) Use of weighting factors from an action spectrum, normalizing an action spectrum, dose rate and dose calculations
3) Use of different sensors to measure surface and underwater UV-A and UV-B radiation, characterization of the chromophoric or colored dissolved organic matter (CDOM) by determination of the concentration of dissolved organic carbon (DOC), and the spectral absorption of the dissolved fraction. (Definitions: immersion effect, cosine response, spectral responsivity, full width half maximum).
4) Determination of mycosporine-like amino acids in planktonic organisms by HPLC
5) Effects of solar UV radiation on aquatic organisms
Spectrophometry
Spectrofluorometry
Radiometry
HPLC
Water organic chemistry (DOC)
Examination is based on an individual report describing the activities during the field and laboratory work.
