It is aim of the lecture to understand drugs, their properties and to derive basic qualities from the structure of a drug substance.

• Structure of drug substances: properties of important functional groups
• Molecular Basis of Drug Action: driving force for drug target interaction, differentiation pharmacodynamics vs. pharmakokinetics, ADME
• Physicochemical properties of drugs: acidity/basicity, drug-like-properties, Lipinski's rule of five
• Biological Membranes: a) structure of biological membranes; b) passage of drugs across membranes: possibilities and structural influences
• Targets: a) types, structure of the most important targets; b) possibilities of drug target interactions
• Syntheses: a) synthesis of basic structures and of selected drugs b) the concept of retrosynthesis
• Stability/chemical instabilities of drugs: a) potentially unstable structures, b) examples for drug instabilities, c) photoinstabilities
• Metabolism: a) General: goals of metabolism, phases, influence of drug application on metabolism, types of reactions, important drug metabolizing enzymes, results of metabolism, toxification and detoxification, influence of the chemical structure; b) examples for drugs metabolism
• Prodrugs: classification, goals, concepts of prodrugs design and clinical applications; most important enzymes involved in prodrug activation b) examples of some important prodrugs
• Discovery of new drugs: a) goals, concepts of drug design, drug development; b) examples
